Prevent people from getting on my computer

It's silly to argue about this. Any security system to anything can be cracked/circumvented/disabled given the right tools and enough time. The point is that there are a a few simple things that you can do that would prevent nearly anyone from even trying to get it.
Well...256bit encryption would take a long long time to crack..its more or less uncrackable.

Yup that is what I was saying for the most part it is dumb considering I have physical access. Now, if he secured his room with a lock that only unlocked via his biometric readings then that would be secure.

Also, encryption is very safe. The NSA has a bounty on anyone who can crack AES encryption. If you can crack it you can claim your 4 billion dollar bounty they are offering on it.

They want to crack it so they can tap into skype conversations since skype uses AES encryption.

So it is pretty damn safe, and if you crack it go claim your 4 billion dollars and then kick out a million to me for letting you know about it.
 
Remove the harddrive, put it in a Faraday Cage bag and lock it in a safe when you're certain you're going to be away from you computer for some time.

Silly, but that's another trick. This can provide the "physical" security along with the logical security of other advices as encryption password, BIOS lockout and basic log-in authentication.
