Running Spotmau from a 64GB USB drive

Hi.
Apparently, Spotmau requires a FAT32 FS, so I'm pretty sure that this is the main issue.

I have a 16GB DOK and all it takes to make it work is connect it and let Spotmau BootSuite "format" it. Basically, the software just quickformats the drive and "burns" an ISO image. After this very short procedure, the drive becomes bootable and Spotmau boots just fine from it.

That's not the case when I do the same on my 64GB USB drive. Well, Spotmau BootSuite does not complain at all. It accepts the media and burns the image just fine. However, immediately after, the drive becomes unreadable under Windows and I'm prompted to format it. Regardless, I tried booting up my PC from it and can you guess what happened?... Yeah, it's a no go.

A while ago I tried several ways of burning the image onto the same drive using different techniques, with no luck. Eventually I had to give up. But now I really need this. The ISO file just sits there on my C drive. How can I burn it onto the 64GB DOK and make it bootable? *Obviously, I need all the available space to be usable (for other stuff) and I really need the NTFS file system, since the 4GB cap on file size is unacceptable.

As a side note: I bought SpotMau awhile back, so what I have is Bootsuite 2012. Is it possible that this problem does not exist in the latest version?
 
If you say so, man. I wouldn't know. I already suspected it to be the problem. So how can I make a bootable NTFS and make SpotMau boot from it? I converted my other (16GB) DOK to NTFS, after letting Spotmau burn its image, and it works and boots just fine! I wish I could do the same with the 64GB drive.
I hope there's a way to make it work. And I really hope it does not require breaking up the drive into several (permanent) partitions.
